The fastest-mutating part of the donor genome is not on the organoid QC panel
A new catalog of tandem repeat variation across seven ape genomes and 69 long-read human and chimpanzee genomes shows these repeats mutate orders of magnitude faster than SNVs, are under strong constraint in coding sequence and 5 prime UTRs, and cluster in nervous-system and synaptic genes. No standard iPSC or organoid quality-control workflow measures them, which makes repeat length a hidden, donor-specific variable in every organoid drug screen.

206 patients in, and tumour-immune organoids still cannot choose your immunotherapy
A PROSPERO-registered systematic review pooled patient-derived tumour-immune organoids as predictors of checkpoint-inhibitor response across 23 studies and 206 patients. Sensitivity was 0.70, the apparent specificity of 0.97 rested on exactly one false positive, every study carried high risk of bias, and the authors conclude the assays should not yet decide whether immunotherapy is added.

Vascularized brain organoids, and the evidence a platform owes its users
Huang and Lin report a co-differentiation protocol that builds endothelial vasculature and microglia-like cells into brain organoids during formation, then apply the same platform to ultrasound-modeled traumatic brain injury and patient-derived glioblastoma. The developmental logic is sound and the molecular concordance is encouraging, but the paper contains no drug-response experiment, no functional activity data, and no reported sample sizes or statistics.

The ECM-exosome growth-plate axis is a testable spec, not a mechanism
A structured narrative review of 46 studies proposes an ECM-exosome axis in the growth plate as a framework for idiopathic short stature. Its own evidence tables show the central interface is untested, and its proposed decisive experiments are, almost verbatim, a requirements document for growth-plate organoids.

The endpoint gap at the heart of organoid-guided cancer trials
NCT03979170 at Geneva University Hospitals proposes 50 lung cancer patients to test whether organoids predict drug response. Its own registry entry shows the registered primary endpoints are organoid establishment and tumor fidelity, not prediction accuracy, and no results have been posted after five years of recruitment.

Osteonecrosis, post-translational modification codes, and the bone organoid gap
A Frontiers review assembles the post-translational modification wiring of osteonecrosis of the femoral head across five cell types. Almost every node was mapped in rodent steroid models or single-cell-type cultures, and the review itself concedes patient-derived femoral head organoids are the missing tool.

EGFR inhibitors turn gut stem cells into hormone cells
Screening more than 600 FDA-approved drugs in human duodenal organoids identified erlotinib and lapatinib as potent inducers of enteroendocrine differentiation, raising chromogranin A-positive cells more than twenty-fold via STAT1 signaling. Lung cancer patients on erlotinib show elevated circulating gut hormones, and treated mice develop enteroendocrine expansion with watery diarrhea, linking an on-target lineage effect to a therapy-limiting side effect.

HIV in a dish: microglia decide what the virus can reach
Cerebral assembloids co-developed from tdTomato-tagged microglial precursors and neural progenitors let HIV infect only the microglia, which then drive interferon and pro-inflammatory signaling into neurons while bystander microglia mount a countervailing neuroprotective program. The model separates injury from adaptation at the earliest stage of brain infection, something bulk-tissue and pre-infection models could not do.

Human taste bud organoids that respond like taste buds
A Seoul and Vienna team derived long-lived organoids from human circumvallate papillae that generate all four taste receptor cell types and respond to bitter, sweet, and umami stimuli with calcium transients. Bitter receptor genotype tracks response, and the stem cell program diverges from mouse, a warning for cross-species taste biology.

EGF versus TGF-alpha: an endocytic switch that sets organoid state
A receptor-proximal RAC1-PLCgamma2 axis sends EGF-bound EGFR into non-clathrin endocytosis while TGF-alpha escapes that route and keeps proliferative signaling alive. Mouse intestinal and mammary organoids expand faster on TGF-alpha, which makes the identity of the growth factor in standard media a genuine confound for drug-response work.

Reading piconewton forces through immune receptors in organoids
Engineered SynNotch receptors convert mechanical force transmitted through CD40 or the T cell receptor into a fluorescent or luminescent readout, working in 2D coculture, in PEG hydrogel lymphoid organoids, and in implanted mice. Because signaling through these receptors scales with the force they bear, force becomes a hidden variable in any immune organoid drug assay.

3'UTR length as a hidden organoid maturation dial
A conserved feedback loop between the neuronal RNA-binding proteins ELAV and Pumilio uses long 3'UTRs to repress translation. Removing one such element from human ELAVL1 causes severe developmental failure in cerebral organoids, revealing post-transcriptional control as a layer most organoid pipelines never measure.

Intrarenal autoantibodies and the organoid immune blind spot
Antibody-mediated rejection of kidney transplants is diagnosed by donor-specific antibodies in blood. This mouse study shows rejecting grafts manufacture their own autoantibodies locally, even when donor-specific antibodies are fully suppressed, and that blocking this in-graft production preserves the organ.

A bankable gut immune co-culture that separates priming from killing
Researchers in Groningen built an autologous human duodenal lymphocyte-organoid co-culture from four pinch biopsies per donor, with bankable immune and epithelial lines and a shared medium that keeps both alive. Its sharpest finding is a negative one: heavy cytokine priming raised every cytotoxicity marker yet killed no epithelium, while only an enforced T cell to epithelium contact trigger did.

Quiescent glioma stem cells hide from the screens built to kill them
Single-nucleus data from ten glioblastomas and patient-derived organoids show about 92 percent of glioma stem cells are slow-cycling or quiescent before treatment, a state that confers radiation resistance but resists deep entry, resembling shallow dormancy. TGF-beta receptor inhibition wakes these cells in 2D and in glioblastoma organoids, while a DYRK1A inhibitor that worked in 2D failed in 3D, a cautionary translation gap for screening programs.

The lung's immune history is written into its organoid assays
In mice, a brief interferon pulse induces a Sca-1-positive alveolar type II cell state that is more proliferative and forms organoids more efficiently, but its post-infection proliferation depends on macrophage-derived oncostatin M. Because Sca-1 has no human ortholog, the finding that organoid-forming efficiency is an inflammation-state readout may not port to human lung models.

Hormone-sensitive stem cells in endometrial organoids
Cells with high aldehyde dehydrogenase activity form endometrial organoids far more efficiently than their low-activity sisters, in mice and in women. Hormones move them between luminal and glandular niches, and their ablation strips glands from the uterus.

Gut lactobacilli prime infant organoids against rotavirus
Ten Lactobacillaceae isolates from human small-intestine donors were screened in infant intestinal organoids. One strain, L. rhamnosus 103, drives IFN-λ secretion through TLR3 via a secreted RNA and restricts replication of the live rotavirus vaccine strain.

Porcine organoids keep regional identity, lose maturity
